CORVALLIS, Ore. Researchers have developed a new drug delivery system that allows inhalation of chemotherapeutic drugs to help treat lung cancer, and in laboratory and animal tests it appears to reduce the systemic damage done to other organs while significantly improving the treatment of lung tumors.

This advance in nanomedicine combines the extraordinarily small size of nanoparticles, existing cancer drugs, and small interfering RNA (siRNA) that shut down the ability of cancer cells to resist attack.

The combination of these forces resulted in the virtual disappearance of lung tumors in experimental animals.

Lung cancer is the leading cancer killer in both men and women. Despite advances in surgery, chemotherapy still plays a major role in its treatment. However, that treatment is constrained by the toxic effects of some drugs needed to combat it and the difficulty of actually getting those drugs into the lungs.

The findings were made by Oleh Taratula at Oregon State University and Tamara Minko and O. Garbuzenko at Rutgers University and the Cancer Institute of New Jersey. They were just published in the Journal of Controlled Release.

"Lung cancer damage is usually not localized, which makes chemotherapy an important part of treatment," said Taratula, an assistant professor in the OSU College of Pharmacy and co-author on this study. "However, the drugs used are toxic and can cause organ damage and severe side effects if given conventionally through intravenous administration.

"A drug delivery system that can be inhaled is a much more efficient approach, targeting just the cancer cells as much as possible," he said. "Other chemotherapeutic approaches only tend to suppress tumors, but this system appears to eliminate it."

A patent is being applied for on the technology, and more testing will be necessary before it is ready for human clinical trials, the researchers said.

The foundation of the new system is a "nanostructured lipid nanocarrier," tiny particles much smaller than a speck of dust that are easily inhaled and also readily attach to cancer cells. This carrier system delivers the anticancer drug. However, it also brings siRNA that makes the cancer cell more vulnerable.

Cancer cells often have two forms of resistance to drugs "pump" resistance that tends to pump the drug out of cells, and "nonpump" resistance that helps keep the cell from dying. The siRNA used in this system helps to eliminate both those forms of resistance, and leaves the cancer cell vulnerable to the drug being used to kill it.

By being inhaled, this system also avoids degradation of the chemotherapeutic agents that occurs when they are injected, researchers said. They arrive in more intact form, ready to do their job on lung cancer cells, while minimizing any side effects.

In more conventional chemotherapy for lung cancer, the drugs tend to accumulate in the liver, kidney and spleen, with much less of the drugs ever making it to the lungs. In this study, the amount of the drug delivered to the lungs rose to 83 percent with the inhalation approach, versus 23 percent with injection.

John Chambers Delivers, Big Investors Now Chasing Stocks

Tuesday, May 21st, 2013
By Mitchell Clark, B.Comm. for Profit Confidential

John Chambers Delivers, Big Investors Now Chasing StocksThe action in the stock market continues to amaze.

When stocks go up on bad news (like last week?s higher initial claims for jobless benefits and lower-than-expected housing starts), you know you don?t want to be short.

Cisco Systems, Inc. (NASDAQ/CSCO) is a component of the Dow Jones Industrial Average, and for such a mature technology stock, it recently reported a very solid quarter.

In its fiscal third quarter (ended April 27, 2012), Cisco announced sales of $12.2 billion, for a net gain of five percent over the comparable quarter. It was the company?s ninth consecutive quarter of record sales.

Earnings grew 14.5% to $2.5 billion, while earnings per share grew 15% to 0.46 per share, beating consensus estimates.

John Chambers, Cisco?s CEO, noted improving signs in the U.S. economy and other markets. Business conditions for the company are also improving.

Like many cash-rich, large corporations, Cisco recently repurchased 41 million shares of its own common stock, spending $860 million.

This, of course, is a pittance. The company finished its latest quarter with cash and cash equivalents of $47.4 billion.

Wall Street boosted the company?s earnings estimates and share price target.

Cisco is ripe for more gains on the stock market because of its valuation.

The company also offers a dividend yield of 3.2%, which is attractive in this market. The company?s huge cash hoard also makes it highly likely that at some point this year, the company will issue another dividend increase.

All institutional investors want to see in this stock market is stability and certainty.

Cisco provided that that certainty in its latest earnings report, and this is the reason why the company has leapt upward by over 10% on the stock market.

Still, the positive trading action is truly astonishing.

If there was ever a case to be made that the stock market is a leading indicator, recent action proves it. This market is way ahead of fundamentals.

Cisco hasn?t done anything for years on the stock market, although it has been slowly growing its revenues.

The company?s latest earnings report really was decent in terms of the competition and maturity in the networking and communication devices subsector.

The broadening of the blue chip breakout into the NASDAQ Composite is confirmation of the uptrend. But it is becoming even more difficult to be a buyer in this stock market when you see the key stock indices going up on bad news.

The appetite that institutional investors have to be buyers is stunning.

A majority of companies beat consensus earnings in the latest quarter. But only half of all companies in the S&P 500 beat consensus on revenues.

This is why big investors are now chasing stocks like Cisco. Top-line growth remains elusive.

CSN: A's Straily outduels Darvish in 1-0 win

BOX SCORE

ARLINGTON, Texas -- Rookie right-hander Dan Straily pitched seven scoreless innings and Yoenis Cespedes homered as the Oakland Athletics won their fifth straight game, 1-0 over the Texas Rangers and Yu Darvish on Tuesday night.

Straily (2-2) gave up only two singles while facing 22 batters, just one over the minimum for his seven innings. He struck out five.

Even with consecutive wins to start the three-game series, Oakland still trails the Rangers by 4 1/2 games in the AL West.

Darvish (7-2) had won all five of his previous starts this season when pitching after a Texas loss.

The Japanese right-hander struck out five, but walked three and hit a batter while giving up five hits in six innings. With the help of a couple of defensive plays, the only run he allowed came when Cespedes homered to straightaway center in the third.

Cespedes' ninth homer went just beyond the reach of Craig Gentry's outstretched glove. Darvish had turned and watched after the slugger took a big swing and connected.

Grant Balfour pitched a perfect ninth for his ninth save in as many chances this season, and his 27th in a row since April 2012, to close out Oakland's third shutout of the season. Sean Doolittle pitched a scoreless eighth for the A's.

Texas was held scoreless for the third time this season.

It was only the 13th 1-0 game ever at Rangers Ballpark, which opened in 1994.

A.J. Pierzynski, the Rangers catcher activated from the disabled list earlier Tuesday, had a one-out single in the third. Jurickson Profar had a single in the sixth, but the rookie was caught stealing when he took off toward second base before Straily threw his pitch.

After winning his season debut April 5 with 11 strikeouts at Houston, Straily was optioned to Triple-A the next day when Bartolo Colon was reinstated from the suspended list. Straily returned in late April, and was 0-2 with an 8.84 ERA in four starts since until dominating the AL West-leading Rangers.

Darvish, who raised his majors-best strikeout total to 91 in 66 2-3 innings, came out after 101 pitches. He had averaged 121 pitches his last three starts, including a career-high 130 five nights earlier against Detroit in a 10-4 victory.

Elvis Andrus made a nifty play to end the fourth, when the shortstop was fully extended when he made a diving stab to catch the ball and then scrambled to his feet and threw out Chris Young at first. Darvish smiled and nodded at Andrus as they made their way to the dugout.

An inning later, after Darvish fielded a bunt by Adam Rosales, the pitcher turned and made a quick throw that went to the left-field side of second base. Andrus caught the ball while stepping on the base, then falling away from the base made a sidearm sling to get Rosales out at first - with a fully extended stretch by second baseman Profar covering the bag.

Notes: Rangers reliever Neal Cotts got three outs on six pitches in his first major league appearance since May 25, 2009. Texas purchased his contract from Triple-A Round Rock earlier Tuesday. The 33-year-old left-hander had ligament replacement surgery in 2009. ... Oakland DH Jed Lowrie left the game with a bruise after fouling a pitch off his right foot. ... 1B Daric Barton, designated for assignment Saturday when A's outfielder Chris Young came off the disabled list, cleared waivers and was outrighted to Triple-A Sacramento. ... Right-hander Chris Resop also cleared waivers and was outrighted to Sacramento.

(AP) ? It was a party atmosphere along Barack Obama's motorcade route from the airport after he arrived in this Central American country for his first visit as president.

Thousands of locals, who are known as "Ticos," lined the road to welcome their fourth visiting American president. The most recent one to visit was Bill Clinton in 1997.

Municipal employees were given the day off to ease traffic.

Some of those who lined the roadway ? two and three people deep ? waved American flags. Others held homemade signs, including one that said "Fired Up!" ? a reference to Obama's campaign slogan. More crowds gathered along the route between Obama's hotel and Casa Amarilla, the headquarters for the Ministry of Foreign Affairs, including hundreds of students in school uniforms.

"We're very happy to welcome our big brother," said retiree Ricardo Marin, 62, explaining the jubilant mood. "We hope they don't neglect us, that this visit will help our country."

Obama's reception here was vastly different from Mexico. There was no major crowd presence for Obama's travels in Mexico City on Thursday and Friday.

___

Excitement over Obama's visit wasn't shared widely, however.

"I had to walk more than a mile to get to my job," grumbled Luis Humberto Rodriguez Burgos, 26, who sells cellphones in the city center, which was shuttered and empty. "There are no clients, no tourists and we're losing money. Clearly if they close the city for a foreign president, that's not going to improve my working conditions, nor help me feed my children."

___

Obama and Costa Rica's president, Laura Chincilla, strolled to a courtyard at Casa Amarilla, or Yellow House, after they met and were greeted by about 100 schoolchildren, ranging in age from about 7 to 16. All were in uniform, and the younger ones waved U.S. and Costa Rican flags. Obama wore a broad grin as he stood encircled by the singing students.

They sang "I'm Tico" and "The First Time I Saw Limon," a reference to a city on the country's Caribbean coast.

As the children sang, thunder roared in the distance and the wind picked up.

___

After the songs, Obama praised the students, calling them "brave" and their performances "very good." He also asked for their names.

The students gushed as Obama invited them to pose for photos.

"What do we say?" he asked.

The students and Chinchilla said: "Cheese." Obama responded with, "Queso!" the Spanish word for cheese.

___

Obama opened his day by making his second visit to Mexico City's soaring, modernistic National Museum of Anthropology.

"Each time that I come, I've been inspired," he told a predominantly student audience.

Of course, last time he may have narrowly avoided getting sick.

In April 2009, Obama was he honored guest at a state dinner at the museum ? just as the global swine flu outbreak was first surfacing in Mexico. Ultimately, the H1N1 strain killed thousands of people worldwide.

Obama never came down with the H1N1 flu. However, a security aide of his did ? and passed it on to family members in the U.S.
